Abstract
The invention provides a method for activating a T lymphocyte by contacting the T lymphocyte with a dendritic cell (DC) that presents a telomerase reverse transcriptase (TRT) peptide in the context of an MHC class I or MHC class II molecule. The DC may be pulsed with a TRT polypeptide or may comprise a recombinant polynucleotide encoding a TRT such as hTRT. The invention also provides DCs comprising a recombinant TRT polynucleotide. The methods and compositions of the invention are used in prevention and treatment of cancers and other cell proliferation diseases or conditions.
Claims23
A method for killing a target cell expressing telomerase reverse transcriptase (TRT), comprising:
- combining human T lymphocytes in vivo with antigen-presenting cells containing a polypeptide that comprises at least 6 consecutive amino acids of telomerase reverse transcriptase (TRT;
- SEQ. ID NO:2), so as to cause T lymphocytes specific for TRT to proliferate;
- and then
- contacting the target cell in vitro with the specific T lymphocytes so as to cause cytolysis of the target cell.
A method for eliciting an anti-TRT immunological response in a human subject, comprising administering to the subject a composition comprising antigen-presenting cells;
- wherein the antigen-presenting cells have been pulsed ex vivo with a synthetic or recombinantly produced polypeptide containing at least 8 consecutive amino acids of telomerase reverse transcriptase (TRT;
- SEQ. ID NO:2), or have been genetically modified with a recombinantly produced polynucleotide encoding said polypeptide.
A method for eliciting an anti-TRT immunological response in a human subject, comprising administering to the subject a composition comprising antigen-presenting cells;
- wherein a nucleic acid preparation comprising nucleic acids that encode at least 30 consecutive amino acids of TRT (SEQ ID NO:2) has been introduced into said antigen presenting cells before administration to the subject.
