Abstract
The present invention is a method for a covalent ligation of one or more molecules to one or more surfaces, that is site-specific and both rapid and high yielding. The covalent ligation to the surface is based on the reaction of an azide and a phosphinothioester to form an amide bond. The method of the invention is particularly well-suited to the immobilization of peptides, proteins or protein fragments to surfaces.
Claims22
A method for immobilizing a molecule on a surface by forming an amide bond which comprises the step of reacting a phosphinothioester with an azide followed by hydrolysis of combined reactants to form an amide bond wherein one of the phosphinothioester or the azide is covalently linked to a surface.
A kit for immobilizing a protein on a surface which comprises a phosphinothiol reagent and a surface to be derivatized with the phosphinothiol reagent.
A kit for immobilizing a protein on a surface, said kit comprising a phosphinothiol reagent, a surface to be derivatized with the phosphinothiol reagent, and one or more reagents for initial derivatization of the surface with a linker.
A surface derivatized with a phosphinothiolester.
