Abstract
Mutants of 2,5-diketo-D-gluconic acid reductase A, an enzyme used to produce 2-keto-L-gulonic acid, a precursor of ascorbic acid (vitamin C) are prepared by site-directed mutagenesis. These mutants have increased catalytic activity, increased expression levels, and/or enhanced temperature stability.
Claims2
An isolated mutant of a 2,5-Diketo-D-Gluconic Acid (2,5-DKG) Reductase A comprising the amino acid sequence of SEQ ID NO:1, wherein said mutant has an increased rate of converting 2,5-DKG to 2-keto-L-gulonic acid compared to the wild-type 2,5-DKG Reductase A of SEQ ID NO:1 and said mutant consists of substitutions at amino acid residues 2, 5, 7, 55, 57 and 192 in SEQ ID NO:1.
