Abstract
The present invention provides improved methods for the production of recombinant peptides from bacterial cells.
Claims21
A method for obtaining a peptide from bacterial cells after expression inside the cells of a fusion protein, wherein the fusion protein comprises the peptide, a carrier protein and an acid-cleavable site between the peptide and the carrier protein, the method comprising:
- treating the bacterial cells for one or more hours with acid under conditions of acid pH and temperature sufficient in a single step to disrupt or lyse the cells and release the peptide from the fusion protein.

- (a) treating the bacterial cells with acid for one or more hours under conditions of acid pH and temperature sufficient to disrupt or lyse the cells and release the peptide from the fusion protein,
- (b) separating soluble material from insoluble material after step (a), and
- (c) recovering the released peptide in the soluble material after step (b).
