Abstract
An Escherichia coli mutant strain deficient in dihydrodipicolinate synthase or dihydrodipicolinate reductase is transformed with a chromosomal gene library of Bacillus methanolicus, and a transformant strain which can grow on a minimal medium is selected. Recombinant DNA which codes for dihydrodipicolinate synthase or dihydrodipicolinate reductase (named dapB) is obtained from the transformant.
Claims12
An isolated DNA which hybridizes under stringent conditions to nucleotides 1 to 798 in SEQ ID NO:
- 3, wherein said stringent conditions comprise washing in 0.1×SSC and 0.1% SDS at 60° C., and wherein said DNA codes for a protein which has dihydrodipicolinate reductase activity.
An isolated DNA which codes for a protein having the amino acid sequence of SEQ ID NO:
- 4.
An isolated DNA which codes for a protein comprising the amino acid sequence of SEQ ID NO:
- 4, which includes substitutions, deletions, insertions, additions, or inversions of 1–10 amino acids, and wherein said protein has dihydrodipicolinate reductase activity.
