Abstract
Novel defective adenoviruses for the transfer and expression of an exogenous nucleotide sequence in a host cell or organism. The invention also relates to novel complementation lines and to the process for the preparation of these novel defective adenoviruses and their use in therapy and to a pharmaceutical composition containing same.
Claims4
An adenoviral vector which is defective for replication, capable of being encapsidated in a complementation cell, and which is derived from an adenoviral genome comprising, from 5′ to 3′, a 5′ ITR, an encapsidation region, an E1A region, an E1B region, an E2 region, an E3 region, an E4 region and a 3′ ITR, by deletion of all or part of the E1A region, all or part of the E4 region, and all of the E3 region, and which comprises a gene of interest placed under the control of the elements needed for its expression in a host cell;
- and further comprising a gene coding for a protein which trans-activates non-adenoviral transcription wherein said gene is placed under the control of the elements needed for the expression of said protein in a host cell.
An adenoviral vector which is defective for replication, capable of being encapsidated in a complementation cell, and which is derived from an adenoviral genome comprising, from 5′ to 3′, a 5′ ITR, an encapsidation region, an E1A region, an E1B region, an E2 region, an E3 region, an E4 region and a 3′ ITR, by deletion of all or part of the E1A region, all or part of the E4 region, and all of the E3 region, and which is, in addition, deleted of a portion of the encapsidation region, and which comprises a gene of interest placed under the control of the elements needed for its expression in a host cell.
