Abstract
Novel parathyroid hormone(PTH) polypeptide derivatives are disclosed, as are pharmaceutical compositions containing said polypeptides, and synthetic and recombinant methods for producing said polypeptides. Also disclosed are methods for treating mammalian conditions characterized by decreases in bone mass using therapeutically effective pharmaceutical compositions containing said polypeptides. Also disclosed are methods for screening candidate compounds of the invention for antagonistic or agonistic effects on parathyroid hormone receptor action. Also disclosed are diagnostic and therapeutic methods of said compounds.

A polypeptide having the amino acid sequence consisting essentially of AlaValAlaGluIleGlnLeuMetHisX01X02X03LysX04 (SEQ ID NO:1), wherein:
- X01 is Ala, Asp or Gln;
- X02 is Leu, Arg or homoArg;
- X03 is Arg or Ala;
- and
- X04 is Phe or Trp;
- and wherein said polypeptide binds to a PTH-1 receptor.
A polypeptide having an amino acid sequence, said sequence consisting essentially of a sequence selected from the group consisting of:
- (a) AlaValAlaGluIleGlnLeuMetHisX01X02X03LysX04(SEQ ID NO:1);
- (b) N- or C- terminal chemical derivatives of said polypeptide having an amino acid sequence of SEQ ID NO:1;
- and
- (c) fragments containing amino acids 1–9, 1–10, 1–11, 1–12 or 1–13 of said polypeptide having an amino acid sequence of SEQ ID NO:1;
- wherein:
- X01 is Ala, Asp or Gln;
- X02 is Leu, Arg or homoArg;
- X03 is Arg or Ala;
- and
- X04 is Phe or Trp;
- and wherein said polypeptide binds to a PTH-1 receptor.
A polypeptide having the amino acid sequence consisting essentially of AlaValAlaGluIleGlnLeuMetHisX01X02X03LysX04 (SEQ ID NO:1), wherein:
- X01 is Ala, Asp or Gln;
- X02 is Leu, Arg or homoArg;
- X03 is Arg or Ala;
- and
- X04 is Phe or Trp;
- and wherein said polypeptide binds to an hDelNt receptor.
A polypeptide having an amino acid sequence, said sequence consisting essentially of a sequence selected from the group consisting of:
- (a) AlaValAlaGluIleGlnLeuMetHisX01X02X03LysX04 (SEQ ID NO:1);
- (b) N- or C- terminal chemical derivatives thereof of said polypeptide having an amino acid sequence of SEQ ID NO:1;
- and
- (c) fragments containing amino acids 1–9, 1–10, 1–11, 1–12 or 1–13 of said polypeptide having an amino acid sequence of SEQ ID NO:1;
- wherein:
- X01 is Ala, Asp or Gln;
- X02 is Leu, Arg or homoArg;
- X03 is Arg or Ala;
- and
- X04 is Phe or Trp;
- and wherein said polypeptide binds to an hDelNt receptor.
