Abstract
The invention relates to methods and materials for use in the typing, diagnosis, prevention and/or treatment of prion disease.

A method for typing PrpSc comprising the steps of:
- (i) providing a first and second sample containing a PrpSc protein;
- (ii) treating the first sample to remove Cu2+ and Zn2+ from PrpSc wherein treatment involves treating the sample with a metal chelator or combination of chelators capable of chelating both Cu2+ and Zn2+;
- (iii) subsequently digesting both the first and second samples with proteinase K;
- and
- (iv) comparing, using side-by-side electrophoresis, the products of the digestion of the first and second samples to determine whether or not the products are the same or different;
- a difference being indicative of the presence of type 1 or type 2 PrpSc, and no difference being indicative of type 3 or type 4 PrpSc.

A method for typing PrpSc comprising:
- (i) providing a sample containing a PrpSc protein;
- (ii) treating the sample to remove Cu2+ and Zn2+ from PrpSc wherein treatment involves treating the sample with a metal chelator or combination of chelators capable of chelating both Cu2+ and Zn2+;
- (iii) subsequently digesting the sample with proteinase K;
- wherein the digestion products of the sample are compared with one or more standard samples of known PrpSc type to identify the type in the sample, a difference being indicative of the presence of type 1 or type 2 PrpSc.
