Abstract
The present invention relates to methods for culturing circovirus and in particular, porcine circovirus. The present invention provides compositions and methods for culturing porcine circovirus in mammalian cells expressing mammalian adenovirus E1 functional protein.
Claims66
A method for culturing a mammalian circovirus comprising:
- a) obtaining mammalian cells expressing a mammalian adenovirus E1 functional protein, wherein said cells are permissive for mammalian circovirus replication;
- b) introducing the mammalian circovirus genome, or a portion thereof capable of replication, into said mammalian cells;
- and
- c) culturing said mammalian cells under conditions suitable for replication of said mammalian circovirus.
A method of preparing a recombinant mammalian cell expressing a mammalian adenovirus E1 functional protein and comprising a porcine circovirus genome comprising the steps of, a) obtaining a mammalian cell expressing a mammalian adenovirus E1 functional protein;
- and b) introducing said porcine circovirus genome, or a portion thereof capable of replication, into the mammalian cell.
A method for replicating a mammalian circovirus comprising, culturing a mammalian cell that comprises a mammalian circovirus genome, or a portion thereof capable of replication, under conditions suitable for replication of said mammalian circovirus, wherein the mammalian cell expresses a mammalian adenovirus E1 functional protein and is permissive for mammalian circovirus replication, and optionally recovering said circovirus from the cultured cell.
A method of preparing a recombinant mammalian cell comprising, introducing a mammalian circovirus genome, or a portion thereof capable of replication, into a mammalian cell that expresses a mammalian adenovirus E1 functional protein, wherein the cell is permissive for replication of the mammalian circovirus.
A method of preparing a recombinant mammalian cell comprising, introducing a mammalian adenovirus E1 gene region into a mammalian cell that comprises a mammalian circovirus genome, or a portion thereof capable of replication, wherein the cell is permissive for replication of the mammalian circovirus.
A method for expressing a mammalian circovirus genome, or portion thereof, comprising, culturing a mammalian cell that comprises the mammalian circovirus genome, or a portion thereof, under conditions suitable for expression of said mammalian circovirus genome, or portion thereof, wherein the mammalian cell expresses a mammalian adenovirus E1 functional protein and optionally recovering said circovirus, or portion thereof, from the cultured cell.
