Abstract
The present invention is directed to nucleotide sequences coding for a bacterial enolase enzyme. These sequences may be used in improved methods for the fermentative preparation of amino acids using coryneform bacteria.
Claims15
A method for production of L-lysine amino acid, comprising:
- a) fermenting a coryneform bacterium that produces said L-lysine amino acid, wherein said coryneform bacterium transformed with a vector comprising an isolated polynucleotide, wherein said isolated polynucleotide is selected from the group consisting of an isolated polynucleotide encoding an enolase enzyme comprising the amino acid sequence of SEQ ID NO:2 or an isolated polynucleotide encoding enolase enzyme comprising the amino acid sequence of SEQ ID NO:2 wherein the glutamic acid at position 223 is replaced with another proteinogenic Lysine acid;
- and
- b) isolating said Lysine amino acid from said coryneform bacteria or from the medium used to grow said coryneform bacterium.
A method for production of L-lysine amino acid, comprising:
- a) fermenting an isolated coryneform bacterium that produces said L-lysine amino acid, wherein said isolated coryneform bacterium comprises a polynucleotide encoding an enolase enzyme comprising the amino acid sequence of SEQ ID NO:2, wherein the glutamic acid at position 223 is replaced by another proteinogenic Lysine amino acid, and
- b) isolating said Lysine amino acid from said coryneform bacteria or from the medium used to grow said coryneform bacterium.
A method for preparing L-lysine amino acid, comprising:
- a) transforming a coryneform bacteria with a vector comprising;
- 1) an isolated polynucleotide encoding enolase enzyme comprising the amino acid sequence of SEQ ID NO:2 or
- 2) an isolated polynucleotide encoding enolase enzyme comprising the amino acid sequence of SEQ ID NO:2, wherein the glutamic acid at position 232 is replaced with another proteinogenic L-lysine amino acid;
- b) culturing said transformed coryneform bacteria under conditions suitable to enhance the activity of said enzyme by overexpression;
- and
- c) isolating the L-lysine amino acid produced.
