Abstract
The present invention provides two new classes of polyene macrolide amide derivatives useful for treating or preventing fungal infections. The new polyene macrolide amide derivatives exhibit antifungal activity and are more water-soluble than conventional polyene antibiotics, such as amphotericin B and amphotericin B methyl ester.
Claims41
A compound according to structural formula (I):
- including pharmaceutically acceptable salts thereof, wherein:
- N—R1—C(O) is a polyene macrolide backbone;
- CH2—R2 is a carbohydrate residue, wherein the illustrated CH2 is derived from the anomeric carbon of a terminal carbohydrate saccharide and R2 represents the remainder of the carbohydrate;
- either (i) R6 is selected from the group consisting of hydrogen, —(CH2)n—NR15R16, —NR15R16, —(CH2)n—R17, polyhydroxylated alkyl, (C1-C6) alkyl substituted with one or more of the same or different R10 group and (C1-C6) heteroalkyl substituted with one or more of the same or different polar substituent, and R7 is selected from the group consisting of —(CH2)n—NR15R16, —NR15R16, —(CH2)n—R17, polyhydroxylated alkyl, (C1-C6) alkyl substituted with one or more of the same or different R10 group, and (C1-C6) heteroalkyl substituted with one or more of the same or different polar substituent;
- or (ii) R6 and R7, taken together with the nitrogen atom to which they are bonded, form 5- or 6-membered saturated or unsaturated ring which optionally includes one or more of the same or different additional heteroatoms selected from the group consisting of O, N, NH, and S and which is optionally substituted at one or more ring carbon or heteroatoms with the same or different substituents selected from the group consisting of a polar substituent, (C1-C6) alkyl, (C1-C6) alkoxy, —(CH2)n—(polar substituent), (C5-C6) aryl, phenyl, 6- to 9-membered arylalkyl and benzyl;
- each R10 is independently selected from the group consisting of ═O (oxo), ═NH (imino), —C(═NH)—NH2 (amidino), and —NH—C(═NH)—NH2 (guanidino);
- R14 is hydrogen or alkyl;
- R15 and R16 taken together with the nitrogen atom to which they are bonded, form a 5- or 6-membered saturated or unsaturated ring which optionally includes one or more of the same or different additional heteroatoms selected from the group consisting of O, N, NH, and S and which is optionally substituted at one or more ring carbon or heteroatoms with the same or different substituents selected from the group consisting of a polar substituent, (C1-C6) alkyl, (C1-C6) alkoxy, —(CH2)n—(polar substituent), (C5-C6) aryl, phenyl, 6- to 9-membered arylalkyl and benzyl;
- R17 is a 5- or 6-membered saturated or unsaturated ring including one or more of the same or different heteroatoms selected from the group consisting of O, N, NH and S and which is optionally substituted at one or more ring carbon or heteroatoms with the same or different substituents selected from the group consisting of polar substituent, (C1-C6) alkyl, (C1-C6) alkoxy, —(CH2)n—(polar substituent), (C5-C6) aryl, phenyl, 6- to 9-membered arylalkyl and benzyl;
- each polar substituent is independently selected from the group consisting of —OH, —SH, ═O, ═S, —NH2, ═NH, —N3, —CN, —C(═NH)—NH2, and —NH—C(═NH)—NH2;
- and
- each n is independently an integer from 1 to 6.
