Abstract
This invention provides methods of generating cells that stably replicate sub-genomic virus replicons. This invention also provides methods of generating cells that have disabled PKR activity and that stably replicate HCV sub-genomic replicons. The invention also provides methods of using the cells of the invention to screen for compounds that modulate viral RNA replication, including HCV RNA replication.
Claims6
A method of screening for compounds that modulate viral replication comprising the steps of
- a) administering a test compound to a cell, wherein said cell comprises a replicating sub-genomic viral replicon, and further wherein said cell is PKR deficient, and
- b) determining whether said test compound modulates the replication of said sub-genomic viral replicon.
A method of screening for compounds that inhibit viral replication comprising the steps of
- a) administering a test compound to a cell, wherein said cell comprises a replicating sub-genomic viral replicon, and further wherein said cell is PKR deficient, and
- b) determining whether said test compound inhibits the replication of said sub-genomic viral replicon.
