Abstract
The present invention concerns an enzymatic oxidative deamination process of a dipeptide monomer to prepare an intermediate useful to prepare compounds having endopeptidase and angiotensin converting enzyme inhibition activity.
Claims29
An isolated DNA sequence encoding the aminotransferase of SEQ ID NO:
- 2, or the complement thereof.
An isolated DNA sequence encoding an amino transferase having at least 80% identity to SEQ ID NO:
- 2, or the complement thereof.
An isolated DNA sequence having the sequence of SEQ ID NO:
- 1 or its complement.
An isolated DNA sequence selected from the group consisting of:
- (a) an isolated DNA sequence encoding a fragment of the amino acid sequence of SEQ ID NO:
- 2 that is catalytically active, or the complement thereof;
- and (b) an isolated DNA sequence capable of hybridizing under stringent conditions to the sequence encoding a fragment of the amino acid sequence of SEQ ID NO:
- 2 that is catalytically active, or the complement thereof, wherein said isolated DNA sequence capable of hybridizing encodes an aminotransferase or a fragment of an aminotransferase that is catalytically active, and wherein said stringent conditions are:
- incubation in 4×SET buffer, 10×Denherdt's solution, 0.1% SDS, 0.1% sodium pyrophosphate, 50 μg/ml denatured salmon sperm DNA at 68° C. for 16-24 hours;
- followed by washing three times for 20 minutes in 3×SET, 0.1% SDS, 0.1% sodium pyrophosphate at 68° C., washing twice for 20 minutes in 1×SET, 0.1% SDS, 0.1% sodium pyrophosphate at 68° C., washing once for 20 minutes in 0.1×SET, 0.1% SDS, 0.1% sodium pyrophosphate at 68° C., and washing once for 20 minutes in 4×SET at room temperature.
An isolated DNA sequence consisting of the sequence of SEQ ID NO:3 or its complement.
