Abstract
Mutagenesis of the gene encoding homoserine dehydrogenase (hom) for production of the amino acid threonine is described. The mutation causes an alteration in the carboxy terminus of the enzyme that interferes with end-product inhibition by threonine. The lack of end-product inhibition causes an overproduction of threonine.
Claims9
A method for the production of threonine comprising
- constructing a gene isolated from a genome selected from the group consisting of Corynebacterium glutamicum, Escherichia coli, and Bacillus subtilis, encoding an enzymatically active homoserine dehydrogenase not subject to allosteric inhibition by threonine, wherein the homoserine dehydrogenase gene is mutated at the carboxy terminus, and
- expressing the gene to prevent end-product inhibition of threonine synthesis.
An enzymaticaly active homoserine dehydrogenase, isolated from bacteria selected from the group consisting of Corynebacterium glutamicum, Escherichia coli, and Bacillus subtillis, not subject to allosteric inhibition by threonine, wherein the enzyme is altered at the carboxy terminus.
