Abstract
Adenoviral mutants are described that have single amino acid mutations in the E1B-55K protein which mutations effect the p53 binding/inactivation and the late functions of the E1B-55K protein in a manner that enhances the efficacy of such viruses for treating cancer when compared to adenoviral mutants that have the E1B-55K region deleted.
Claims7
A recombinant adenovirus comprising a mutation in the E1B-55K gene, said gene encoding a mutated E1B-55K protein comprising a single amino acid mutation, said single amino acid mutation reducing the ability of said E1B-55K mutated protein to bind to the tumor suppressor p53 when compared to the wild-type E1B-55K protein and said adenovirus has the further property of retaining late viral function.
A method of restoring adenoviral E1B55K mutants'0 replication cold insensitivity, comprising the steps of mutating the E1B region of said mutant and assaying for cold insensitive replication, and selecting those adenoviral mutants that are replication cold insensitive.
