Abstract
A method of detecting a target nucleotide sequence in a nucleic acid molecule, which comprises: (a) binding of an oligonucleotide probe to said nucleic acid molecule; (b) selective labelling of the bound oligonucleotide probe in the presence of said target nucleotide sequence; (c) hybridization of the labelled oligonucleotide to a complementary sequence; and (d) subsequent detection of the label; such methods being suitable for qualitative and quantitative assays of microbiological populations.

A method of detecting a target nucleotide sequence in a nucleic acid molecule which comprises:
- (a) binding of an oligonucleotide probe to said nucleic acid molecule to provide a complex between the oligonucleotide probe and nucleic acid molecule;
- (b) selective labeling of the bound oligonucleotide probe in the presence of said target nucleotide sequence;
- (c) denaturation of the complex between the oligonucleotide probe and nucleic acid molecule;
- (d) incubation of the labeled and any unlabeled oligonucleotide and a complementary sequence under conditions whereby labeled oligonucleotide hybridizes to the complementary sequence;
- and
- (e) subsequent detection of the label wherein detection of the label is indicative of detection of the target nucleotide sequence.
A method of determining the amount of a target nucleotide sequence in a nucleic acid molecule or the number of cells containing a target nucleotide sequence in a nucleic acid molecule which comprises
- (a) binding of an oligonucleotide probe to said nucleic acid molecule to provide a complex between the oligonucleotide probe and nucleic acid molecule;
- (b) selective labeling of the bound oligonucleotide probe in the presence of said target nucleotide sequence;
- (c) denaturation of the complex between the oligonucletoide probe and nucleic acid molecule;
- (d) incubation of labeled and any unlabeled oligonucleotide and a complementary sequence under conditions whereby labeled oligonucleotide hybridizes to the complementary sequence;
- (e) subsequent detection of the label;
- and
- (f) quantification of the label, wherein the quantity of the label is correlated with the amount of the target sequence or the number of cells containing the target sequence.
