Abstract
Described herein are methods that can be used for diagnosis and prognosis of cellular proliferation. Also described herein are methods that can be used to screen candidate bioactive agents for the ability to modulate cellular proliferation. Additionally, methods and molecular targets (genes and their products) for therapeutic intervention in cancers are described.
Claims41
A method of screening for a bioactive agent capable of binding to a domain of cellular proliferation protein KSP (SEQ ID NO:2), wherein said domain of said cellular proliferation protein (i) is KSP-L360 (amino acids 1-360 of SEQ ID NO:4), KSP-K491 (amino acids 1-491 of SEQ ID NO:6), KSP-S553 (amino acids 1-553 of SEQ ID NO:8), KSP-K368 (SEQ ID NO:10), or a fragment or a variant thereof, said variant having greater than 80% amino acid sequence identity to said domain of said cellular proliferation protein and (ii) has microtubule binding activity, said method comprising:
- (a) combining in vitro said domain of said cellular proliferation protein and a candidate bioactive agent, and
- (b) determining the binding of said candidate bioactive agent to said domain of said cellular proliferation protein, provided that said candidate bioactive agent is not a microtubule, ATP, or an antibody.
A method for screening for a bioactive agent capable of modulating the ATPase activity of a domain of cellular proliferation protein KSP (SEQ ID NO:2), wherein said domain of said cellular proliferation protein (i) is KSP-L360 (amino acids 1-360 of SEQ ID NO:
- 4), KSP-K491 (amino acids 1-491 SEQ ID NO:6), KSP-S553 (amino acids 1-553 of SEQ ID NO:8), KSP-K368 (SEQ ID NO:10), or a fragment or variant thereof, said variant having greater than 80% amino acid sequence identity with said domain of said cellular proliferation protein, and (ii) has ATPase activity, said method comprising:
- (a) combining in vitro said domain of said cellular proliferation protein and a candidate bioactive agent;
- and
- (b) determining the effect of said candidate bioactive agent on the ATPase activity of said domain of said cellular proliferation protein, provided that said candidate bioactive agent is not a microtubule, ATP, or an antibody.
A method for determining the effect of a bioactive agent on a cell, said method comprising:
- (a) combining in vitro a domain of cellular proliferation protein KSP (SEQ ID NO:2) and a candidate bioactive agent, wherein said domain of said cellular proliferation protein (i) is KSP-L360 (amino acids 1-360 of SEQ ID NO:4), KSP-K491 (amino acids 1-491 of SEQ ID NO:6), KSP-S553 (amino acids 1-553 of SEQ ID NO:8), KSP-K368 (SEQ ID NO:
- 10), or a fragment or a variant thereof, said variant having greater than 80% amino acid sequence identity with said domain of said cellular proliferation protein, and (ii) has ATPase activity or microtubule binding activity;
- (b) determining the binding of said candidate bioactive agent to said domain of said cellular proliferation protein to identify a bioactive agent that binds to said domain, provided that said candidate bioactive agent is not a microtubule, ATP, or an antibody;
- (c) providing a cell that expresses recombinant human KSP (SEQ ID NO:
- 2), or a fragment thereof that has ATPase activity or microtubule binding activity;
- (d) contacting said bioactive agent identified in step (b) with said cell under conditions in which the bioactive agent is taken up by the cell;
- and
- (e) determining whether said bioactive agent has an effect on said cell, wherein determining the effect on said cell comprises determining the effect of said bioactive agent on cellular proliferation, on cellular viability, on cellular morphology, or on a mitotic spindle.
A method for determining the effect of a bioactive agent on a cell, said method comprising:
- (a) combining in vitro a domain of cellular proliferation protein KSP (SEQ ID NO:2) and a candidate bioactive agent, wherein said domain of said cellular proliferation protein (i) is KSP-L360 (amino acids 1-360 of SEQ ID NO:4), KSP-K491 (amino acids 1-491 of SEQ ID NO:6), KSP-S553 (amino acids 1-553 of SEQ ID NO:8), KSP-K368 SEQ ID NO:10), or a fragment or a variant thereof, said variant having greater than 80% sequence identity with said domain of said cellular proliferation protein, and (ii) has ATPase activity;
- (b) determining the effect of said candidate bioactive agent on the ATPase activity of said domain of said cellular proliferation protein to identify a bioactive agent that modulates the ATPase activity of said domain, provided that said candidate bioactive agent is not a microtubule, ATP, or an antibody;
- (c) providing a cell that expresses recombinant human KSP (SEQ ID NO:2), or a fragment thereof that has ATPase activity or microtubule binding activity,
- (d) contacting the bioactive agent identified in step (b) with said cell under conditions in which the bioactive agent is taken up by the cell;
- and
- (e) determining whether said bioactive agent has an effect on said cell, wherein determining the effect on said cell comprises determining the effect of said bioactive agent on cellular proliferation, on cellular viability, on cellular morphology, or on a mitotic spindle.
