Abstract

The present invention relates to an vitro device for the removal of ionized substances from a membrane sample without mechanical penetration, which device comprises: (a) a positive electrode; (b) a negative electrode, and (c) electrical insulation between subpart (a) and (b), wherein the positive electrode, and the negative electrode, and electrical insulation are positioned on the same side of the membrane sample. The present invention also relates to a device for the removal of or delivery of ionized substances to a mammal through intact skin or mucosal membrane without mechanical penetration, which device comprises: (a) a positive electrode, (b) a negative electrode, and (c) an electrically insulating material between subpart (a) and (b), wherein the positive electrode, negative electrode and insulating material are physically positioned so that each present a common surface of the device for contact with the same surface of the skin or mucosal membrane of the mammal. The present invention also relates to the use of iontophoresis to determine the level of a charged molecule in a living mammal, and with the use of a feedback mechanism, administer appropriate keels of therapeutic substances.

Dependent claims

A method of sampling a substance present in a subject, said method comprising:

(a) placing a first collection reservoir comprising an electrically conducting medium in contact with a skin surface of said subject, wherein said first reservoir is in electrical contact with a first electrode;
(b) placing a second collection reservoir comprising an electrically conducting medium in contact with a separate skin surface of said subject, wherein said second reservoir is in electrical contact with a second electrode;
(c) sampling the substance by establishing a potential between said first and second electrodes such that detectable levels of said substance and/or at least one of its metabolites is extracted into said first and second collection reservoirs;
and
(d) measuring the level of said extracted substance and/or metabolites present in each of said first and second collection reservoirs to obtain first and second measurements.

A monitoring system for use in sampling of glucose at physiological pH from a mammal through an intact skin or mucosal membrane of said mammal and detection of said glucose, said system comprising:

(1) an iontophoretic device comprising:
(A) electrical leads for a positive electrode and a negative electrode, said leads connected to a power supply and capable of creating an electrical circuit;
(B) means for correlating a signal from extracted glucose with an amount or concentration of glucose;
and
(C) a display to indicate said amount or concentration of glucose in operative communication with said means for correlating;
(2) a component for use in said device, comprising:
(A) a positive electrode, a negative electrode, and an electrically insulating material separating the electrodes, wherein:
(i) the positive electrode, negative electrode and insulating material are physically positioned to present a single common surface, and
(ii) said positive and negative electrodes are in operative communication with said electrical leads;
(B) a first collection reservoir comprising an electrically conductive material positioned to be capable of electrical communication with said negative electrode;
and
(C) a second collection reservoir comprising an electrically conductive material, wherein said second collection reservoir is positioned to be capable of electrical communication with said positive electrode;
and
(3) an analyzing component in operative communication with said means for correlating.
