Abstract
The invention concerns a novel method for purifying and quantifying viral particles. More particularly, the invention concerns a method for purifying and quantifying adenovirus by ion-exchange chromatography. The invention also concerns a method for identifying different adenovirus serotypes.
Claims14
A method of separating viral particles from a biological medium selected from the group consisting of a supernatant of encapsidation cells producing the virus particles and a lysate of encapsidation cells producing the viral particles, the method comprising contacting the biological medium with a support comprising a cross-linked agarose matrix and ion-exchange groups bound to the cross-linked agarose matrix by a flexible arm, such that contact between the biological medium and the support chromatographically separates the viral particles from the biological medium.
A method of quantifying adenoviruses in a supernatant or lysate of an adenovirus encapsidation culture, comprising the steps of:
- contacting the supernatant or lysate with a support in order to chromatographically separate the adenoviruses from the supernatant or lysate,
- measuring the absorbance of the adenoviruses separated from the supernatant or lysate, and
- calculating from the measured absorbance the amount or concentration of the adenoviruses separated from the supernatant or lysate;
- wherein
- the support comprises a cross-linked 6% agarose matrix with quaternary amine groups covalently attached to the matrix by way of flexible, hydrophilic dextran arms having a mean molecular weight of about 5000 Da, wherein the matrix has a particle size of between 45 and 165 μm and a mean particle size of 90 μm.
