Abstract
DNA encoding human neuronal nicotinic acetylcholine receptor alpha and beta subunits, mammalian and amphibian cells containing said DNA, methods for producing alpha and beta subunits and recombinant (i.e., isolated or substantially pure) alpha subunits (specifically α6) and beta subunits (specifically β3) are provided. In addition, combinations of a plurality of subunits (i.e., one or more of as α1, α2, α3, α4, α5, α6 and/or α7 subunits in combination with one or more of β3 subunits are provided.
Claims43
An isolated DNA molecule, comprising a sequence of nucleotides that encode an α6-subunit of a human neuronal nicotinic acetylcholine receptor, wherein the sequence of nucleotides encoding the α6 subunit is selected from the group consisting of:
- a) a sequence of nucleotides that encodes a human α6 subunit and comprises the coding portion of the sequence of nucleotides as set forth in SEQ ID NO:
- 1;
- b) a sequence of nucleotides that encodes a human α6 subunit that comprises the sequence of amino acids as set forth in SEQ ID NO:
- 2;
- and
- c) a sequence of nucleotides which is degenerate as a result of the genetic code to the sequence of nucleotides as set forth in any of (a) or (b).
An isolated DNA molecule, comprising a sequence of nucleotides that encode a full length β3 subunit of a human neuronal nicotinic acetylcholine receptor, wherein the sequence of nucleotides encoding the β3 subunit is selected from the group consisting of:
- a) a sequence of nucleotides that encodes a human β3 subunit and comprises the coding sequence of nucleotides as set forth in SEQ ID NO:
- 3;
- b) a sequence of nucleotides that encodes a human β3 subunit that comprises the sequence of amino acids as set forth in SEQ ID NO:. 4;
- and
- c) a sequence of nucleotides which is degenerate as a result of the genetic code to the sequence of nucleotides as set forth in any of (a) or (b).
An isolated DNA molecule, comprising a sequence of nucleotides that encodes amino acids 1-28 of SEQ. ID No. 4.
An isolated DNA molecule, comprising a sequence of nucleotides that encodes amino acids. 1-20of SEQ ID. No. 4.
An isolated DNA molecule, comprising nucleotides 98-202 set forth in SEQ ID No. 3.
