Abstract
One aspect of the present invention relates to novel heterocyclic compounds comprising urea functionality. A second aspect of the present invention relates to the use of the novel heterocyclic compounds comprising urea functionality as ligands for various cellular receptors, including opioid receptors, other G-protein-coupled receptors and ion channels. An additional aspect of the present invention relates to the use of the novel heterocyclic compounds comprising urea functionality as analgesics.
Claims8
A compound represented by structure A:
- wherein
- X represents CH2;
- Z represents O, S or NR;
- R represents independently for each occurrence H, C1-C10 alkyl, aryl or monocyclic or bicyclic heteroaryl with 5-12 ring atoms, of which one to three ring atoms are selected independently from the group consisting of S, O, and N;
- R1 represents H, C1-C10 alkyl, alkenyl, alkynyl, aryl, monocyclic or bicyclic heteroaryl with 5-12 ring atoms, of which one to three ring atoms are selected independently from the group consisting of S, O, and N, C1-C20 aralkyl or monocyclic or bicyclic heteroaralkyl with 5-12 ring atoms, of which one to three ring atoms are selected independently from the group consisting of S, O, and N;
- R2 represents independently for each occurrence H, C1-C10 alky, alkenyl, alkynyl, aryl, C1-C20 aralkyl or alkyl substituted with monocyclic or bicyclic heteroaryl with 5-12 ring atoms, of which one to three ring atoms are selected independently from the group consisting of S, O, and N, or the two instances or R2 taken together represent an aryl or monocyclic or bicyclic heteroaryl with 5-12 ring atoms, of which one to three ring atoms are selected independently from the group consisting of S, O, and N, or a —CH2—, —CH2CH2—, —CH2CH2CH2—, or —CH2CH2CH2CH2—tether between the two nitrogens of the urea moiety;
- R3 represents H, C1-C10 alkyl, C1-C10 fluoroalkyl, alkenyl, alkynyl, C1-C20 aralkyl or alkyl substituted with monocyclic or bicyclic heteroaryl with 5-12 ring atoms, of which one to three ring atoms are selected independently from the group consisting of S, O, and N;
- m is an integer in the range 1 to 4 inclusive;
- n is equal to 1;
- p is an integer in the range 1 to 3 inclusive;
- and
- the stereochemical configuration at any stereocenter of a compound represented by A may be R, S, or a mixture of these configurations.
