Abstract
A novel transcription enhancer element (AATT)n, that increases the activity of cis-elements in heterologous promoter constructs without altering their intrinsic specificity is provided. Also provided is a transcription factor, PABF, which specifically binds to the (AATT) repeat element and methods of use therefor.
Claims13
A method of increasing transcription of an endogeous gene in a cell comprising:
- transfecting a cell comprising an endogenous gene controlled by an (AATT)n repeat element with a nucleic acid encoding a palindromic element binding factor (PABF), wherein said PABF is characterized as;
- a) having a molecular weight of approximately 67 kD, as dcterrnined by SDS-PAGE, and b) hatting an H1 histone domain a glutamine rich domain, and an HMG/Y domain;
- incubating said cell under conditions to express said PABF;
- and
- detecting increased transcription of said endogenous gene in said cell.

A method of increasing transcription of an exogenous Gene comprising:
- transfecting a cell with a first nucleic acid comprising an (AATT)n repeat element functionally linked to a promoter and an exogenous gene:
- and
- transfecting said cell with a second nucleic acid encoding a palindromic binding factor (PABF), wherein said PABF is characterized as:
- a) having a molecular weight of approximately 67 kD, as determined by SDS-PAGE, and b) having an H1 histone domain, a glutamine rich domain, and an HMG/Y domain, whereby transcription of said exogenous gene is increased relative to transcription from a nucleic acid lacking said (AATT)n repeat element.
