Abstract
Isolated polypeptides useful in ameliorating GAD-associated autoimmune disease as well as diagnostic and therapeutic methods of using the peptides are disclosed.

A method for determining the status of a GAD associated autoimmune disorder in a patient which comprises contacting T-cells of said patient with at least one peptide that is selected from the group consisting of:
- (a) KPCSCSKVDVNYAFLHATDL (SEQ ID NO:06);
- (b) NMYAMMIARFKMFPEVKEKG (SEQ ID NO:07);
- (c) TAGTTVYGAFDPLLAVADICKK (SEQ ID NO:08);

- (e) IPPSLRYLEDNEERMSRLSK (SEQ ID NO:10);

- and

A method for detecting a GAD associated autoimmune disorder in a patient having or at risk of having the disorder, said method comprising contacting T-cells from said patient with a diagnostically effective amount of a diagnostically effective fragment of GAD65 and detecting the response of said T-cells to said fragment of GAD65 wherein said T-cell response is indicative of the presence of said disorder, wherein said diagnostically effective fragment is selected from the group consisting of:

A method for staging a β-cell associated autoimmune disease of a patient suspected of having a susceptibility to said autoimmune disease, said method comprising:
- determining the autoreactivity of T-cells of said patient toward at least one peptide that is selected from the group consisting of:
- (a) KPCSCSKVDVNYAFLHATDL (SEQ ID NO:06);
- (b) NMYAMMIARFKMFPEVKEKG (SEQ ID NO:07);
- (c) TAGTTVYGAFDPLLAVADICKK (SEQ ID NO:08);
- (d) EYLYNIIKNREGYEMVFDGK (SEQ ID NO:09);
- (e) IPPSLRYLEDNEERMSRLSK (SEQ ID NO:10);
- (f) SRLSKVAPIKARMMEYGTT (SEQ ID NO:11);
- (g) EYGTTMVSYQPLGDKVNFFR (SEQ ID NO:12);
- and

A kit useful for determining the status of a GAD associated disorder in a specimen of a patient, said kit comprising one or more containers comprising a polypeptide that is IPPSLRYLEDNEERMSRLSK (SEQ ID NO:10).
