Abstract
The present invention relates to methods and compositions for enhancing cardiac function in mammalian hearts by inserting transgenes that increase β-adrenergic responsiveness within the myocardium. The present invention can thus be used in the treatment of heart disease, especially congestive heart failure.
Claims27
A method of enhancing the cardiac function of a mammal with congestive heart failure comprising administering a replication-deficient adenoviral vector comprising a nucleic acid sequence encoding a cardiac adenylylcyclase (AC) protein operatively linked to a promoter, to at least one coronary artery of the mammal such that the cardiac AC protein is expressed in the myocardium and enhances the cardiac function of the mammal.
A method of enhancing the cardiac function of a mammal with congestive heart failure comprising administering a replication-deficient adenoviral vector comprising a nucleic acid sequence encoding a cardiac adenylylcyclase (AC) protein operatively linked to a promoter, to at least one saphenous vein graft or mammary artery graft supply blood to the myocardium of the mammal such that the AC protein is expressed in the myocardium and enhaces the cardiac function of the mammal.
The method of clalim 15, wherein the enhanced cardiac function of the mammal is left ventricular dP/dt.
