Abstract
The present invention relates to a novel ppGpp synthetase and expression systems for improved production of proteins of interest in Gram-positive bacteria, involving use of the novel ppGpp synthetase.
Claims9
A method for producing a protein of interest, which comprises:
- (a) culturing a gram-positive bacterial strain, wherein (i) a DNA sequence encoding the protein of interest has been introduced into the strain, (ii) the strain produces less of a polypeptide having ppGpp synthetase activity than the corresponding wild-type strain, and (iii) the polypeptide having ppGpp synthetase activity has an amino acid sequence that is at least 70% identical to the amino acid sequence of amino acid residues 1 to 734 of SEQ ID NO:2;
- and
- (b) purifying the protein of interest from the resulting culture broth or expression system.
