Abstract
The present invention is directed to a composition and method for regulating the adhesion of cells and biomolecules to hydrophobic surfaces and hydrophobic coated surfaces. The composition is a biomolecule conjugated end-group activated polymer (FGAP). The biomolecule conjugated EGAP can be put to numerous uses including cell adhesion, cell growth, cell sorting, and other biological assays.

A method for the attachment of cells or viruses to a surface for growth or biological analysis comprising the steps of:
- a. obtaining a PEO- and PPO-containing triblock or diblock copolymer, wherein said copolymer comprises an end group;
- b. activating the end group of said copolymer;
- c. contacting a hydrophobic surface with said copolymer for a time sufficient for the copolymer to be adsorbed by the hydrophobic surface;
- d. conjugating a thiol containing biomolecule to said copolymer to form a biomolecule-conjugated copolymer-coated surface;
- e. contacting the biomolecule-conjugated copolymer-coated surface with a cell or virus such that said cell or virus adheres to the biomolecule-conjugated copolymer-coated surface.
A method for the attachment of cells for growth or biological analysis comprising the steps of:
- a. contacting a hydrophobic surface with a PEO- and PPO-containing triblock or diblock copolymer comprising an activated end group for a time sufficient for said coplymer to be adsorbed by the hydrophobic surface;
- b. conjugating a natural or recombinant biomolecule to the end group of the copolymer adsorbed to the hydrophobic surface to form a biomolecule-conjugated copolymer-coated surface;
- c. contacting said biomolecule-conjugated copolymer-coated surface with at least one cell such that said cell adheres to the biomolecule-conjugated copolymer-coated surface.
A method of selecting at least one desired cell or virus from a mixture of at least two cells or viruses comprising the steps of:
- a. contacting a hydrophobic surface with a PEO- and PPO-containing triblock or diblock copolymer comprising an activated end group for a time sufficient for said copolymer to be adsorbed by the surface;
- b. conjugating a biomolecule to the end group of said surface-attached copolymer to yield a biomolecule-conjugated copolymer-coated surface, said biomolecule being unique for the desired cell or virus being selected;
- c. contacting the biomolecule-conjugated copolymer-coated surface with a mixture of cells or viruses containing the desired cell or virus;
- d. allowing the desired cell or virus to adhere to the biomolecule-conjugated copolymer-coated surface;
- and
- e. removing the non-adhered cells or viruses.
A method for the attachment of cells to a surface for growth or biological analysis such that said cells may be released from the surface by treatment with a thiol-containing reagent, wherein said method comprises the steps of:
- a. contacting a hydrophobic surface with a PEO- and PPO-containing triblock or diblock copolymer comprising a disulfide derivative group for a time sufficient for said copolymer to be adsorbed by the hydrophobic surface;
- b. forming a disulfide linkage between a natural or recombinant biomolecule comprising a thiol group and the disulfide derivative group of the copolymer adsorbed to the hydrophobic surface to form a biomolecule-conjugated copolymer-coated surface;
- c. contacting said biomolecule-conjugated copolymer-coated surface with at least one cell such that said at least one cell adheres to the biomolecule-conjugated copolymer-coated surface.
