Abstract
Disclosed are methods, nucleic acids, and cells for expressing an exogenous gene in a mammalian cell, involving introducing into the cell a non-mammalian DNA virus (e.g., a baculovirus) whose genome carries an exogenous gene, and growing the cell under conditions such that the gene is expressed. Also disclosed are methods for treating gene deficiency disorders, neurological disorders, or cancers in a mammal by providing to a cell a therapeutically effective amount of a virus whose genome carries an exogenous, therapeutic gene and growing the cell under conditions such that the exogenous gene is expressed in the mammal.

A method of expressing an exogenous gene in a mammalian cell, said method comprising:
- a) introducing into the cell a nuclear non-mammalian DNA virus or an entomopoxvirus, other than the prokaryote virus the genome of which comprises said exogenous gene under the control of a promoter that induces expression of the exogenous gene in the cell to produce an infected cell;
- and
- b) maintaining the infected cell under conditions such that said exogenous gene is expressed.
A nucleic acid comprising:
- a genome of a nuclear non-mammalian DNA virus or an entomopoxvirus;
- an exogenous mammalian gene;
- and
- an exogenous mammalian-active promoter operably linked to said exogenous gene.
A nucleic acid comprising:
- a genome of a nuclear non-mammalian DNA virus or an entomopoxvirus;
- an exogenous antisense RNA gene, the RNA encoded by said gene being complementary to a nucleic acid of a gene that is expressed in a cell at an undesirably high level;
- and
- an exogenous mammalian-active promoter operably linked to said exogenous antisense RNA gene.
A non-human cell that contains a nucleic acid, wherein said nucleic acid comprises:
- a genome of a nuclear non-mammalian DNA virus or an entomopoxvirus;
- an exogenous mammalian gene;
- and
- an exogenous mammalian-active promoter operably linked to said exogenous gene.
A nucleic acid comprising:
- a genome of a nuclear non-mammalian DNA virus or an entomopoxvirus;
- an exogenous cancer therapeutic gene selected from the group consisting of tumor necrosis factor genes, thymidine kinase genes, chimeric diphtheria toxin genes, and cytosine deaminase genes;
- and
- an exogenous mammalian-active promoter operably linked to said exogenous cancer therapeutic gene.
A nucleic acid comprising:
- a genome of a nuclear non-mammalian DNA virus or an entomopoxvirus;
- an exogenous gene selected from the group consisting of RNA decoy genes and ribozyme genes;
- and
- an exogenous mammalian-active promoter operably linked to said exogenous gene.
A cell in vitro that contains a nucleic acid, wherein said nucleic acid comprises:
- a genome of a nuclear non-mammalian DNA virus or an entomopoxvirus;
- an exogenous mammalian gene;
- and
- an exogenous mammalian-active promoter operably linked to said exogenous mammalian gene.
