Abstract
A support for solid phase amplification or sequencing of nucleic acids has a functionalized solid support, a linker arm having functional groups covalently bound to the solid support through at least one binding site of the functional groups, and an oligonucleotide primer bound at its 5′ end to the linker, the primer, thereby, being immobilized on the solid support.
Claims19
A support for the solid phase amplification or sequencing of nucleic acids comprising:
- a polyfunctionalized solid support;
- a linker having a multiplicity of functional groups capable of binding to said solid support;
- and
- an oligonucleotide primer bound at a 5′ end to said linker;
- wherein said primer is immobilized on said solid support via said linker by means of at least one covalent bond between said functional groups and said solid support.
The support of claim, wherein said linker i is a polymer molecule.
A method for preparing a support for solid phase amplification or sequencing of nucleic acids, comprising:
- providing a linker covalently bond to a 5′ end of an oligonucleotide primer, said linker having functional groups which provide a multiplicity of potential binding sites;
- and contacting an amount of said linker with a polyfunctionalized solid support under reaction conditions whereby various covalent bonds are formed between at least one of said potential binding sites of each linker and said solid support.
