Abstract
A method of eliciting an immune response against an antigen in a vertebrate subject, the method comprising the steps of providing an antigen-adjuvant composition comprising the antigen and a substantially non-toxic adjuvant molecule having biological activity in mucosal tissues, and administering said antigen-adjuvant composition to the vertebrate subject in a manner such that initial contact occurs in mucosal tissue of the vertebrate subject, whereby an immune response is elicited. Cytokines are preferred adjuvants. Preferred cytokines are interleukin-1α (IL-1α) and interleukin-1β (IL-1β).

A method of eliciting an immune response against an antigen in a vertebrate subject, the method comprising the steps of:
- (a) providing an antigen-adjuvant composition comprising the antigen and a substantially nontoxic cytokine adjuvant molecule having biological activity in mucosal tissues and having less immunogenicity than cholera toxin, wherein the antigen-adjuvant composition is soluble in water, and wherein the cytokine adjuvant molecule is selected from the group consisting of an IL, GM-CSF, and combinations thereof;
- and
- (b) administering said antigen-adjuvant composition intramucosally to the vertebrate subject in a manner such that initial contact occurs in mucosal tissue of the vertebrate subject, whereby an immune response is elicited, the immune response comprising a systemic immune response.
A method of eliciting an immune response against an antigen in a vertebrate subject, the method comprising the steps of:
- (a) providing an antigen-adjuvant composition comprising the antigen and a cytokine adjuvant molecule having biological activity in mucosal tissues, wherein the antigen-adjuvant composition is free of alum, wherein the antigen-adjuvant composition is soluble in water, wherein the cytokine adjuvant molecule is selected from the group consisting of an IL, GM-CSF and combinations thereof, and wherein the antigen and adjuvant are not conjugated together;
- and
- (b) administering said antigen-adjuvant composition intramucosally to the vertebrate subject in a manner such that initial contact occurs in mucosal tissue of the vertebrate subject, whereby an immune response is elicited, the immune response comprising a systemic immune response.
A method of eliciting an immune response against an antigen in a vertebrate subject, the method comprising:
- (a) providing a water-soluble antigen-adjuvant composition comprising the antigen and IL-1β;
- and
- (b) administering said antigen-adjuvant composition intramucosally to the vertebrate subject in a manner such that initial contact occurs in mucosal tissue of the vertebrate subject, whereby an immune response is elicited.
