Abstract
Isolated polynucleotide molecules provide RSV genome and antigenomes, including that of human, bovine or murine RSV or RSV-like viruses, and chimera thereof. The recombinant genome or antigenome can be expressed with a nucleocapsid (N) protein, a nucleocapsid phosphoprotein (P), a large (L) polymerase protein, and an RNA polymerase elongation factor to produce isolated infectious RSV particles. The recombinant RSV genome and antigenome can be modified to produce desired phenotypic changes, such as attenuated viruses for vaccine use.
Claims22
An isolated infectious, self-replicating RSV particle which comprises a recombinant RSV genome or antigenome, a nucleocapsid (N) protein, a nucleocapsid phosphoprotein (P), a large (L) polymerase protein, and a M2(ORF1) RNA polymerase elongation factor.
A transfected, transformed, or transduced cell comprising one or more recombinant expression vector(s) that encode a RSV genome or antigenome and N, P, L and M2(ORF1) RNA polymerase elongation factor proteins of RSV.
A composition which comprises one or more isolated polynucleotide molecule(s) comprising at least one operably linked transcriptional promoter, at least one polynucleotide sequence that encode(s) an RSV genome or antigenome and N, P, L and M2(ORF1) RNA polymerase elongation factor proteins of RSV, and a transcriptional terminator.
