Abstract
Viruses expressing ligands on their surfaces are used as a detection means for the related polypeptide which binds the ligand. Multiple copies of the ligand can be expressed on the viral surface. These viruses may be used to detect polypeptides, cells, receptors and channel proteins.
Claims25
A method of isolating a cell expressing a polypeptide on its surface, comprising the steps of:
- contacting a sample of cells with a population of virus, wherein each virus expresses on its surface at least 10 copies of a ligand previously demonstrated to be specific for the polypeptide expresssed on the surface of the cell;
- and
- isolating bound virus, thereby isolating a cell expressing the polypeptide on its surface.
A method of isolating a cell expressing a polypeptide on its surface, comprising the steps of:
- contacting a sample of cells with a population of bactcriophage, wherein each bacteriophage expresses on its surface at least 10 copies of a ligand previously demonstrated to be specific for the polypeptide on the surface of the cell;
- and
- isolating bound bacteriophage, thereby isolating a cell expressing the polypeptide on its surface.
A method of isolating a cell expressing a polypeptide on its surface comprising:
- contacting a sample of cells with a population of virus, wherein each virus expresses on its surface at least 10 copies of a polypeptide ligand specific for the polypeptide on the surface of the cell, to form a mixture of cells and virus, whereby a subset of the cells in the sample bind to the virus;
- and
- isolating bound virus from the mixture, thereby isolating cells which are bound to the virus by virtuc of expressing the polypeptide on the cell surface.
A method of isolating a cell expressing a polypeptide on its surface comprising:
- contacting a sample of cells with a population of bacteriophage, wherein each bacteripbage expresses on its surface at least 10 copies of a polypeptide ligand specific for the polypeptide on the surface of the cell, to form a mixture of cells and bacteriophage, whereby a subset of the cells in the sample bind to the bacteriophage;
- and
- isolating bound bacteriophage from the mixture, thereby isolating cells which are bound to the bacteriophage by virtue of expressing the polypeptide on the cell surface.
