Abstract
Method and compositions for regulating cell cycle progression are disclosed. Compositions include nucleic acids comprising a human Cdc5 gene, antisense gene and fragments thereof and a human Cdc5 protein and polypeptide fragments thereof polypeptide. The compositions can be used in a method to treat diseases relating to a cell cycle defect.
Claims18
An isolated nucleic acid comprising the sequence of FIG. 2D (SEQ ID No:
- 11).
An isolated nucleic acid encoding the hCdc5 protein comprising the sequence of SEQ ID No:
- 1.
An isolated oligonucleotide probe for hCdc5, comprising at least 15 consecutive nucleotides of SEQ ID NO:
- 11 or its complement, wherein the oligonucleotide probe is a member of an oligonucleotide probe pair for amplification of a hCdc5 sequence.
An antisense nucleic acid comprising a nucleic acid sequence complementary to the nucleic acid sequence of FIG. 2D (SEQ ID NO:
- 11).
method of inhibiting translation of the mRNA of a human Cdc5 gene by a target cell comprising contacting said target cell with an oligonucleotide probe, wherein said probe comnrises at least 15 consecutive nucleotides of the complement of SEQ ID NO:
- 11, under conditions in which said oligonucleotide probe hybridizes to said mRNA, whereby translation of said mRNA is inhibited, wherein said contacting of said target cell occurs in vitro.
A method for detecting a human Cdc5 gene in a sample, said method comprising:
- contacting said sample with an oligonucleotide probe for the hCdc5 gene, wherein said probe comprises at least 15 consecutive nucleotides of the sequence of SEQ ID No:
- 11, or its complement, under conditions in which said probe hybridizes to said human Cdc5 gene, and
- detecting the presence of the hybridized probe.
A method for detecting a hCdc5 mRNA in a sample, said method comprising:
- contacting said sample with an oligonucleotide probe for the hCdc5 gene, wherein said probe comprises at least 15 consecutive nucleotides of the complement of the sequence of SEQ ID No:
- 11 under conditions in which said probe hybridizes to said hCdc5 mRNA, and
- detecting the presence of the hybridized probe.
An isolated oligonucleotide probe for hCdc5, comprising at least 15 consecutive nucleotides of the coding region of SEQ ID NO:
- 11, or the complement thereof.
