Abstract
Transgenic swine in which the normal expression of .alpha.(1,3) galactosyltransferase is prevented in at least one organ of tissue type. The absence or inactivation of this enzyme prevents the production of carbohydrate moieties having the distinctive terminal Gal.alpha.1-3Gal.beta.1-4GlcNAc epitope that is a significant factor in xenogeneic, particularly human, transplant rejection of swine grafts.
Claims5
Porcine cells, in vitro, said cells comprising a disrupted .alpha.(1,3)-galactosyltransferase gene encoding the polypeptide of SEQ.ID.NO:1, whereby expression of functional .alpha.(1,3)-galactosyltransferase is inhibited.
A process for producing a modified porcine cell, comprising:
- treating a porcine cell, in vitro, said treating introducing a disruption into the .alpha.(1,3)-galactosyltransferase gene encoding the polypeptide of SEQ.ID.NO:1, whereby expression of functional .alpha.(1,3)-galactosyltransferase is inhibited.
