Abstract
The invention provides hybrid oligonucleotides having phosphorothioate or phosphorodithioate internucleotide linkages, and both deoxyribonucleosides and ribonucleosides or 2'-substituted ribonucleosides. Such hybrid oligonucleotides have superior properties of duplex formation with RNA, nuclease resistance, and RNase H activation.
Claims53
A hybrid antisense oligonucleotide that is complementary to a single stranded target nucleic acid and inhibits expression of the target nucleic acid, the hybrid antisense oligonucleotide comprising at least four contiguous deoxyribonucleotides and at least two 2'-O-methyl-substituted ribonucleotides at the 3'-end of the oligonucleotide, and further comprising one or more phosphorothioate internucleotide linkages.
A hybrid antisense oligonucleotide that is complementary to a single stranded target nucleic acid and inhibits expression of the target nucleic acid, the hybrid antisense oligonucleotide comprising at least three contiguous deoxyribonucleotides and at least two 2'-O-alkoxyalkyl-substituted ribonucleotides at the 3'-end of the oligonucleotide, and further comprising one or more phosphorothioate internucleotide linkages.
