Abstract
Expression plasmids comprising a nucleic acid sequence of interest controlled by a T7 bacteriophage promoter and a stabilizing region comprising all or part of the par region of the RP4 plasmid or a derivative thereof. The invention also concerns the use of said plasmids in the production of recombinant products.
Claims19
An expression plasmid comprising a nucleic acid sequence encoding a protein of interest under the control of a bacteriophage T7 promoter and a stabilizing region comprising all or part of the par region of the plasmid RP4, or of a derivative of the par region wherein said all or part or derivative of the par region of the plasmid RP4 stabilizes the plasmid such that the plasmid may be maintained in bacterial cells without any selection pressure.
A plasmid pXL2435 comprising SEQ ID NO:1.
A process for preparing recombinant aFGF, said process comprising culturing a bacterium comprising plasmid pXL2435 and a gene encoding the bacteriophage T7 RNA polymerase under conditions which produce said aFGF.
