Abstract
The invention provides a primary liver stem cell and a cell doublet consisting of a hepatocyte and the stem cell, both of which are derived from normal liver tissue. Methods of isolating the cells, genetically altering the cells, and using the cells for transplantation are also within the invention.
Claims50
An isolated liver cell cluster of less than 10 cells comprising a liver stem cell and a hepatocyte, wherein said stem cell does not express OC2.
A primary liver stem cell wherein said stem cell is
- obtained from normal liver tissue, and
- does not express OC2.
A method of obtaining an isolated liver stem cell comprising
- (a) isolating a liver cell cluster of less than 10 cells from normal liver tissue, said cluster comprising an OC2-negative stem cell associated with a hepatocyte;
- (b) dissociating said stem cell from said hepatocyte;
- and
- (c) removing said hepatocyte from said cluster to yield a liver stem cell.
A primary liver stem cell, wherein said stem cell
