Abstract
Polypeptides that are cleared from the kidney and do not contain in their original form a Fc region of an IgG are altered so as to comprise a salvage receptor binding epitope of an Fc region of an IgG and thereby have increased circulatory half-life.
Claims14
A polypeptide which is cleared from the kidney and does not contain a Fc region of an IgG, altered to comprise a salvage receptor binding epitope of an Fc region of an IgG, wherein the epitope comprises the sequence PKNSSMISNTP (SEQ ID NO:
- 3).
A polypeptide variant of a polypeptide of interest which is cleared from the kidney, wherein the polypeptide of interest is a Fab or F(ab').sub.2 which contains an Ig domain or an Ig-like domain comprising at least one of a C.sub.H 1 or C.sub.L region, wherein the polypeptide of interest does not contain a Fc region of an IgG, in which the variant varies from the polypeptide of interest by comprising a salvage receptor binding epitope of an Fc region of an IgG, wherein the epitope is taken from a loop of a C.sub.H 2 domain, wherein the loop consists of PKNSSMISNTP (SEQ ID NO:
- 3) and wherein the variant has a longer in vivo half-life than the polypeptide of interest.
