Aromatic C16-C20 substituted tetrahydro prostaglandins useful as FP agonists

A method of treating a human or other animal subject having a bone disorder, said method comprising administering to said subject a compound according to the structure:

• ##STR39## wherein (a) R.sub.1 is CO.sub.2 H, C(O)NHOH, CO.sub.2 R.sub.5, CH.sub.2 OH, S(O).sub.2 R.sub.5, C(O)NHR.sub.5, C(O)NHS(O).sub.2 R.sub.5, or tetrazole;
• wherein R.sub.5 is alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• (b) R.sub.2 is H or lower alkyl;
• (c) X is NR.sub.6 R.sub.7, OR.sub.8, SR.sub.9, S(O)R.sub.9, or S(O).sub.2 R.sub.9 ;
• wherein R.sub.6, R.sub.7, and R.sub.8 are independently selected from the group consisting of H, acyl, alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• and wherein R.sub.9 is alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• (d) R.sub.3 and R.sub.4 are independently H, alkyl, hydroxyalkyl, alkoxyalkyl, OR.sub.10, or SR.sub.10 except that both R.sub.3 and R.sub.4 are not H and when one of R.sub.3 and R.sub.4 is H, the other is not methyl;
• wherein R.sub.10 is alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring, R.sub.10 having from 1 to about 8 member atoms;
• (e) Z is carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• and
• any optical isomer, diastereomer, enantiomer of the above structure, or a pharmaceutically-acceptable salt, or biohydrolyzable amide, ester, or imide thereof.

A method of treating glaucoma, said method comprising administering to a human or other animal a safe and effective amount of a compound according to the structure:

• ##STR40## wherein (a) R.sub.1 is CO.sub.2 H, C(O)NHOH, CO.sub.2 R.sub.5, CH.sub.2 OH, S(O).sub.2 R.sub.5, C(O)NHR.sub.5, C(O)NHS(O).sub.2 R.sub.5, or tetrazole;
• wherein R.sub.5 is alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• (b) R.sub.2 is H or lower alkyl;
• (c) X is NR.sub.6 R.sub.7, OR.sub.8, SR.sub.9, S(O)R.sub.9, or S(O).sub.2 R.sub.9 ;
• wherein R.sub.6, R.sub.7, and R.sub.8 are independently selected from the group consisting of H, acyl, alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• and wherein R.sub.9 is alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• (d) R.sub.3 and R.sub.4 are independently H, alkyl, hydroxyalkyl, alkoxyalkyl, OR.sub.10, or SR.sub.10, except that both R.sub.3 and R.sub.4 are not H and one of R.sub.3 and R.sub.4 is H, the other is not methyl;
• wherein R.sub.10 is alkyl, heteroalkyl, carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring, R.sub.10 having from 1 to about 8 member atoms;
• (e) Z is carbocyclic aliphatic ring, heterocyclic aliphatic ring, aromatic ring, or heteroaromatic ring;
• and any optical isomer, diastereomer, enantiomer of the above structure, or a pharmaceutically-acceptable salt, or biohydrolyzable amide, ester, or imide thereof.
