Abstract
The present invention provides compositions and methods for transfecting eukaryotic cells which comprise a nucleic acid, a cationic lipid capable of forming a complex with said nucleic acid, and a viral agent which is an active or inactive enveloped virus or a component of an enveloped virus. Particular viral agents include alpharviruses and components thereof.
Claims97
A composition for transfecting a eukaryotic cell with a nucleic acid which comprises a nucleic acid, a cationic lipid composition which comprises a cationic lipid and a viral agent which is an enveloped virus or a component of an enveloped virus with the exception that the viral component is not influenza virus hemagglutinin and wherein said nucleic acid is not a nucleic acid of said viral agent.
A composition for transfecting a eukaryotic cell with a nucleic acid which comprises a nucleic acid, a cationic lipid composition which comprises a polyvalent cationic lipid and a viral agent which is an enveloped virus or a component of an enveloped virus and wherein said nucleic acid is not a nucleic acid of said viral agent.
A composition for transfecting a eukaryotic cell with a nucleic acid which comprises a nucleic acid, a cationic lipid composition and a viral agent which is an enveloped virus or a component of an enveloped virus and said enveloped virus is a togavirus or a rhabdovirus and said nucleic acid is not a nucleic acid of said viral agent.
A composition for transfecting a eukaryotic cell with a nucleic acid which comprises a nucleic acid, a cationic lipid composition and a viral agent which is an alphavirus virus or a component of an alphavirus and wherein said nucleic acid is not a nucleic acid of said viral agent.
A composition for transfecting a eukaryotic cell with a nucleic acid which comprises a nucleic acid, a cationic lipid composition and a viral agent which comprises a mixture of fusion peptides from at least two different enveloped viruses.
A composition for transfecting a eukaryotic cell with a nucleic acid which comprises a nucleic acid, a cationic lipid composition and a viral agent is a vesicular stomatitus virus or a component thereof and wherein said nucleic acid is not a nucleic acid of a vesicular stomatitus virus.
A composition for facilitating introduction of a biologically-active anionic macromolecule into a eukaryotic cell which comprises a cationic lipid composition and a viral agent which is a viral agent selected from the group consisting of a togavirus, a rhabdovirus, an alphavirus, a Sindbis virus, a Semliki Forest virus, a Vesicular Stomatitis virus, components thereof and a mixture of two or more fusion peptides of an enveloped virus.
A method for making a composition for transfecting a eukaryotic cell with a nucleic acid which comprises the steps of:
- combining a cationic lipid composition comprising a polyvalent cationic lipid, a viral agent which is an enveloped virus or a component of an enveloped virus and a nucleic acid wherein said nucleic acid is not a nucleic acid of said viral agent, and
- forming a cationic liposome or vesicle from said cationic lipid composition.
