Abstract
Disclosed and claimed are a method for the covalent attachment of poly- and oligosaccharides to protein molecules via hydrogen peroxide depolymerization of the polysaccharide units, followed by attachment of the depolymerized polysaccharide chain to the amino acid groups of a protein of interest through a linker molecule, and products therefrom, and methods for using the products.
Claims18
A method for preparing a construct comprising a polysaccharide and/or oligosaccharide covalently attached to a protein molecule, said method comprising
- depolymerizing and activating said polysaccharide and/or oligosaccharide using hydrogen peroxide under mild conditions, wherein said mild conditions include reaction at a temperature from about 30.degree. C. to about 80.degree. C. and a pH from about 4.5 to about 8.0, and wherein no metal catalysts are added;
- derivatizing the depolymerized polysaccharide and/or oligosaccharide with a molecule selected from the group consisting of an amine and a hydrazide, wherein the derivatization is carried out in the presence of a carbodiimide;
- and
- conjugating said derivatized, depolymerized polysaccharide and/or oligosaccharide with a protein molecule.
