Abstract
Compounds and methods are provided for use in purification of CD34.sup.+ cells and specific surface antigens thereof. The present invention discloses methods for releasing CD34.sup.+ cells, as well as compounds having a carbohydrate epitope of the CD34 surface antigen, from an affinity matrix, using carbohydrates having the structure: Neu5Ac.alpha.2-3Gal.beta.1-4(X) wherein (X) is GlcNAc, or a monosaccharide or a cyclohexane derivative that is structurally similar to GlcNAc.
Claims10
A method for screening for a candidate compound able to bind to CD34.sup.+ cells, comprising the steps of:
- (a) contacting a candidate compound with an isolated carbohydrate having the structure:
- Neu5Ac.alpha.2-3Gal.beta.1-4(X)
- wherein (X) is GlcNAc, or a monosaccharide or a cyclohexane derivative that is structurally similar to GlcNAc;
- and
- (b) detecting candidate compound bound to said carbohydrate, thereby identifying a candidate compound that binds to CD34.sup.+ cells.
A method for screening for a candidate compound able to bind to CD34.sup.+ cells, comprising the steps of:
- (a) contacting a candidate compound with an isolated carbohydrate having the structure:
- Neu5Ac.alpha.2-3Gal.beta.1-4(X)
- wherein (X) is GlcNAc, or a monosaccharide or a cyclohexane derivative that is structurally similar to GlcNAc, and wherein said carbohydrate is immobilized on a solid support;
- (b) contacting said immobilized carbohydrate with a binding partner for said carbohydrate;
- (c) separating unbound binding partner from said immobilized carbohydrate;
- and
- (d) detecting binding partner bound to said immobilized carbohydrate, thereby identifying a candidate compound that binds to CD34.sup.+ cells.
