Abstract
A purified and isolated DNA sequence and the encoded mammary-specific secreted protein, mammaglobin, are disclosed. Also disclosed are methods for detecting breast cancer based upon the overexpression and secretion of mammaglobin by breast cancer cells. The methods detect and/or quantitate the presence of mammaglobin or the mRNA encoding mammaglobin. Immunotherapy-based methods for treating a breast cancer patient with a mammaglobin-expressing tumor are also disclosed. The methods involve using mammaglobin antigens to induce a humoral and/or cell-mediated immune response against the tumor.
Claims13
An isolated and purified polypeptide comprising at least one mammaglobin antigen which comprises at least twelve contiguous amino acids of SEQ ID NO:2, wherein said mammaglobin antigen is recognized by B cells or T.sub.C cells, or by both B cells and T.sub.C cells, said B cells and T.sub.C cells being specific for a naturally occurring, secreted mammaglobin polypeptide which is glycosylated.
An isolated and purified polypeptide comprising at least one mammaglobin T.sub.C cell antigen which comprises at least six contiguous amino acids of SEQ ID NO:2, wherein said mammaglobin T.sub.C cell antigen is recognized by T.sub.C cells specific for a naturally occurring, secreted mammaglobin polypeptide which is glycosylated.
An isolated and purified polypeptide comprising at least one mammaglobin B cell antigen which comprises at least six contiguous amino acids of SEQ ID NO:2 and a T.sub.H cell epitope from a heterologous protein, wherein said mammaglobin B cell antigen is recognized by B cells specific for a naturally occurring, secreted mammaglobin polypeptide which is glycosylated.
