Abstract
The present invention discloses multivalent single chain antibodies which have two or more biologically active antigen binding sites. The multivalent single chain antibodies are formed by using a peptide linker to covalently link two or more single chain antibodies, each single chain antibody having a variable light domain linked to a variable heavy chain domain by a peptide linker.
Claims4
A multivalent single chain antibody which comprises two or more single chain antibodies, each single chain antibody specifically binding TAG-72, wherein the single chain antibodies are covalently linked together by at least one first peptide linker which comprises a 15-25 amino acid segment of the amino acid sequence (SEQ ID NO:
- 5), Leu-Ser-Ala-Asp-Asp-Ala-Lys-Lys-Asp-Ala-Ala-Lys-Lys-Asp-Asp-Ala-Lys-Lys-As p-Asp-Ala-Lys-Lys-Asp-Leu
- and each single chain antibody comprises
- (a) a first polypeptide comprising a light chain variable domain;
- (b) a second polypeptide comprising a heavy chain variable domain;
- and
- (c) a second peptide linker linking die first and second polypeptides into a functional binding moiety.
