Abstract
The invention relates to the production of AAV capsids which may be used to transfer native or heterologous molecules into appropriate host cells. The capsid proteins can be expressed from a recombinant virus, expression vector, or from a cell line that has stably integrated the AAV capsid genes or coding sequences. The invention further provides for the production of AAV capsids in vitro from the AAV capsid proteins and the construction of packaged capsids in vitro. The invention further provides for the production of AAV capsids that have been genetically engineered to express heterologous epitopes of clinically important antigens to elicit an immune response.
Claims17
An adeno-associated virus capsid vehicle comprising adeno-associated virus capsid proteins covalently linked, bound to, or encapsidating an RNA molecule.
An adeno-associated virus capsid vehicle comprising adeno-associated virus capsid proteins covalently linked, bound to, or encapsidating a protein.
An adeno-associated virus capsid vehicle comprising adeno-associated virus capsid proteins covalently linked, bound to, or encapsidating a polypeptide.
An adeno-associated virus capsid vehicle comprising adeno-associated virus capsid proteins covalently linked, bound to, or encapsidating a small organic molecule.
An adeno-associated virus capsid vehicle, comprising an adeno-associated virus capsid protein engineered to contain a heterologous epitope.
A method for in vitro assembly of an adeno-associated virus capsid vehicle, comprising:
- (a) recovery of AAV capsid proteins from cells expressing capsid proteins;
- (b) combining the AAV capsid proteins with a molecule to be bound to, or covalently linked to the AAV capsid proteins, thereby resulting in the assembly of an AAV capsid vehicle.
A method for in vitro assembly of an adeno-associated virus capsid vehicle, comprising:
- (a) recovery of AAV capsid proteins from cells expressing capsid proteins;
- (b) combining the AAV capsid proteins under conditions for reassembly with a molecule to be packaged within the capsid vehicle.
