Abstract
A method for dissolving crystalline L-alpha-aspartyl-L-phenyl alanine methyl ester product is disclosed. The product is obtained by crystallizing the ester from an aqueous solution, by cooling. The initial concentration of ester in the aqueous solution used provides at least 10 grams of precipitated solid phase per liter of solution. The solution is cooled through conductive heat transfer without effecting forced flow to form a sherbet-like pseudo solid phase
Claims22
A method of dissolving L-.alpha.-aspartyl-L-phenylalanine methyl ester, comprising dissolving in water, L-.alpha.-aspartyl-L-phenylalanine methyl ester which had been crystallized by cooling, without effecting forced flow, an industrial-scale aqueous solution of L-.alpha.-aspartyl-L-phenylalanine methyl ester containing an initial concentration of said L-.alpha.-aspartyl-L-phenylalanine methyl esters sufficient to obtain about 10 grams or more of precipitated solid phase per liter of solvent after cooling, wherein said solution is cooled by conductive heat transfer to form a sherbet-like pseudo solid phase.
An industrial-scale method of dissolving L-.alpha.-aspartyl-L-phenylalanine methyl ester, comprising dissolving L-.alpha.-aspartyl-L-phenylalanine methyl ester obtained by a process comprising:
- (i) adjusting the initial concentration of L-.alpha.-aspartyl-L-phenylalanine methyl ester in industrial-scale aqueous solution thereof to obtain about 10 grams or more of precipitated solid phase per liter of solvent after cooling;
- and
- (ii) cooling said solution through conductive heat transfer without causing forced flow by mechanical stirring to form a sherbet-pseudo solid phase.
An industrial-scale method of dissolving L-.alpha.-aspartyl-L-phenylalanine methyl ester, comprising dissolving L-.alpha.-aspartyl-L-phenylalanine methyl ester obtained by a process comprising the steps of:
- (i) preparing an industrial-scale aqueous solution of L-.alpha.-aspartyl-L-phenylalanine methyl ester having an initial concentration of said L-.alpha.-aspartyl-L-phenylalanine methyl ester in said solution to obtain about 10 grams or more of precipitated solid phase per liter of solvent after cooling;
- (ii) charging said solution into an industrial crystallizer equipped with solid cooling surfaces having a refrigeration medium circulated therein and comprising cooling plates or cooling tubes, and a cooling jacket, to cool said solution without effecting forced flow and by conductive heat transfer, keeping a distance between said solution and said solid cooling surfaces of 500 mm or less to form a sherbet-like pseudo solid phase;
- (iii) isolating said sherbet-like pseudo solid phase;
- and
- (iv) drying said sherbet-like pseudo solid phase to obtain said crystalline L-.alpha.-aspartyl-L-phenylalanine methyl ester.
