Abstract
This invention relates to a purified Helicobacter pylori vacuolating toxin and methods to use this toxin to produce protective antibodies against H. pylori infection. Antiserum to this antigen can be used to detect the toxin. Methods to detect anti-toxin antibodies determine the susceptibility of a patient to develop peptic ulcer disease, gastric carcinoma, or other clinical consequences of H. pylori infection.
Claims7
An isolated nucleic acid encoding an H. pylori vacuolating toxin, or a nucleic acid fragment consisting of at least 20 nucleotides from an H. pylori vacuolating toxin-encoding nucleic acid.
An isolated nucleic acid encoding an H. pylori vacuolating toxin comprising the nucleic acid set forth in the Sequence Listing as SEQ ID NO:1.
An isolated nucleic acid consisting of the nucleotides set forth in the Sequence Listing as SEQ ID NO:1.
An isolated oligonucleotide consisting of the nucleotides set forth in the Sequence Listing as SEQ ID NO:21.
An isolated oligonucleotide consisting of the nucleotides set forth in the Sequence Listing as SEQ ID NO:22.
A diagnostic test kit for detecting H. pylori toxin nucleic acids in a sample, said test kit comprising:
- (a) two single stranded oligonucleotides of at least 20 nucleotides consisting of nucleic acid fragments specific for the nucleic acid set out in sequence Id. no 1;
- (b) means for conducting PCR reaction;
- and
- (c) means for detecting H. pylori toxin nucleic acids.
