Abstract
The invention relates to a compound of the formula I ##STR1## wherein R.sup.1 is a cyclic imide group and X, Y, R.sup.2, and R.sup.3 are as defined herein. The invention further relates to pharmaceutical compositions containing, and methods of using, compounds of the formula I. Compounds of the formula I are useful in the treatment of diseases related to the production of matrix metalloproteinases and tumor necrosis factor.
Claims11
A compound of the formula ##STR5## wherein X is CH.sub.2 or NH;
- Y is a bond, CH.sub.2, CH((C.sub.1 -C.sub.6)alkyl), CF.sub.2, S, or O;
- R.sup.1 is an optionally substituted cyclic imide wherein the cyclic imide ring system has the formula ##STR6## in which A, B, C and D are each CH, and E and F each independently represent CH and where the optional substituents in formulas (i), (ii), (iii), and (iv) are halogen, hydroxyl, phenyl, (C.sub.1 -C.sub.6)alkyl, (C.sub.1 -C.sub.6)alkoxy, or (C.sub.1 -C.sub.6)alkylthio;
- R.sup.2 is hydrogen, (C.sub.1 -C.sub.6)alkyl, (C.sub.6 -C.sub.10)aryl, (C.sub.5 -C.sub.9)heteroaryl, (C.sub.5 -C.sub.9)heteroaryl(C.sub.1 -C.sub.6)alkyl, (C.sub.6 -C.sub.10)aryl(C.sub.1 -C.sub.6)alkyl, (C.sub.1 -C.sub.6)alkylthio(C.sub.1 -C.sub.6)alkyl, (C.sub.1 -C.sub.6)alkylsulfinyl(C.sub.1 -C.sub.6)alkyl, (C.sub.1 -C.sub.6)alkylsulfonyl(C.sub.1 -C.sub.6)alkyl, (C.sub.3 -C.sub.6)cycloalkyl!(C.sub.1 -C.sub.4)alkyl or (C.sub.1 -C.sub.6)alkyl substituted by a substituent selected from the group consisting of hydroxy, amino, (C.sub.1 -C.sub.6)alkylamino, ((C.sub.1 -C.sub.6)alkyl).sub.2 amino, --COOR.sup.4, and --CONR.sup.5 R.sup.6, wherein R.sup.4, R.sup.5 and R.sup.6 are each independently selected from the group consisting of hydrogen, (C.sub.1 -C.sub.6)alkyl, (C.sub.6 -C.sub.10)aryl(C.sub.1 -C.sub.6)alkyl, and (C.sub.5 -C.sub.9)heteroaryl(C.sub.1 -C.sub.6)alkyl;
- and,
- R.sup.3 is hydrogen, (C.sub.6 -C.sub.10)aryl, (C.sub.5 -C.sub.9)heteroaryl, (C.sub.1 -C.sub.6)alkyl, (C.sub.6 -C.sub.10)aryl(C.sub.1 -C.sub.6)alkyl, (C.sub.5 -C.sub.9)heteroaryl(C.sub.1 -C.sub.6)alkyl,piperidyl(C.sub.1 -C.sub.6)alkyl,N-((C.sub.1 -C.sub.6)alkyl)piperidyl(C.sub.1 -C.sub.6)alkyl, or (C.sub.2 -C.sub.6)alkyl substituted by hydroxy, piperazino, amino, ((C.sub.1 -C.sub.6)alkyl)amino, ((C.sub.1 -C.sub.6)alkyl).sub.2 amino, morpholino, thiomorpholino, piperidino, pyrrolidino, N-acylpiperazino, N-((C.sub.1 -C.sub.6)alkyl)piperazino, N-((C.sub.6 -C.sub.10)aryl)piperazino, N-((C.sub.5 -C.sub.9)heteroaryl)piperazino,N-((C.sub.6 -C.sub.10)aryl(C.sub.1 -C.sub.6)alkyl)piperazino,N-((C.sub.5 -C.sub.9)heteroaryl(C.sub.1 -C.sub.6)alkyl)piperazino, (C.sub.1 -C.sub.6)alkylthio, sulfinyl or sulfoxyl.
