Abstract
A method for the production of monoclonal antibodies in mammal's milk through the creation of transgenic animals that selectively express foreign antibody genes in mammary epithelial cells.
Claims10
A transgenic non-human mammal all of whose germ cells and somatic cells contain a heterologous immunoglobulin protein-coding sequence operatively linked to a promoter sequence that directs the preferential expression of said protein-coding sequence in mammary gland epithelial cells, thereby providing a heterologous and assembled immunoglobulin in the milk of said mammal wherein said heterologous and assembled immunoglobulin is in a functional configuration and is produced at levels of at least about 1 mg/ml in the milk of said mammal.
A transgenic non-human goat all of whose germ cells and somatic cells contain a heterologous immunoglobulin protein-coding sequence operatively linked to a promoter sequence that directs the preferential expression of said protein-coding sequence in mammary gland epithelial cells, thereby providing a heterologous and assembled immunoglobulin in the milk of said goat, wherein said heterologous and assembled immunoglobulin is in a functional configuration and is produced at levels of at least about 1 mg/ml in the milk of said goat.
