Abstract
The invention relates to a process for producing an immunogenic construct comprising activating at least one first carbohydrate-containing moiety with CDAP, CTEA or pNPC, and covalently joining the activated first moiety to a second moiety. Preferably, the first moiety is a polysaccharide and the second moiety is a protein. Immunogenic constructs are prepared by this process using either direct or indirect conjugation of the first and second moieties.
Claims24
A method of producing an immunogenic construct, comprising:
- (a) activating at least one first carbohydrate-containing moiety selected from the group consisting of polysaccharides, oligosaccharides, disaccharides, and monosaccharides with an organic cyanylating reagent selected from the group consisting of 1-cyano-4-(dimethylamino)-pyridinium tetrafluoroborate, N-cyanotriethyl-ammonium tetrafluoroborate, and p-nitrophenylcyanate, to form an activated carbohydrate;
- (b) derivatizing a second moiety selected from the group consisting of proteins, peptides, and haptens with a thiol or hydrazide nucleophile group;
- and
- (c) coupling said activated carbohydrate directly or indirectly to the derivatized second moiety to form an immunogenic construct capable of stimulating an immune response.
A method for producing an immune response, comprising:
- (a) preparing an immunogenic construct by (i) activating at least one first carbohydrate-containing moiety selected from the group consisting of polysaccharides, oligosaccharides, disaccharides, and monosaccharides with an organic cyanylating reagent selected from the group consisting of 1-cyano-4-(dimethylamino)-pyridinium tetrafluoroborate, N-cyanotriethyl-ammonium tetrafluoroborate, and p-nitrophenylcyanate, (ii) derivatizing a second moiety selected from the group consisting of proteins, peptides, and haptens with a thiol or hydrazide nucleophile group, and (iii) covalently joining said activated carbohydrate directly or indirectly to the derivatized second moiety;
- and
- (b) administering the immunogenic construct to a patient.
