Abstract
The invention provides substituted-2-amino-1,2,3,4-tetrahydrodibenzofurans and 2-aminocycloheptab!benzo-furans useful as 5-HT.sub.1F agonists.
Claims9
A compound of the Formula I ##STR19## wherein:
- R.sup.1 and R.sup.2 are independently hydrogen, C.sub.1 -C.sub.4 alkyl, benzyl, or .alpha.-methyl-4-nitrobenzyl;
- X is nitro, halo, --OH, --NH.sub.2, --CN, --NHC(O)R.sup.3, --C(O)R.sup.6, --NHSO.sub.2 R.sup.7, or --SO.sub.2 NHR.sup.10 ;
- R.sup.3 is C.sub.1 -C.sub.6 alkyl, C.sub.2 -C.sub.6 alkenyl, C.sub.3 -C.sub.8 cycloalkyl, phenyl, substituted phenyl, naphthyl, or phenyl(C.sub.1 -C.sub.4 alkylene);
- R.sup.6 is hydroxy, amino, C.sub.1 -C.sub.6 alkoxy, benzyloxy, phenoxy, or --NHR.sup.8 ;
- R.sup.7 is C.sub.1 -C.sub.6 alkyl, phenyl or phenyl monosubstituted with halo or C.sub.1 -C.sub.4 alkyl;
- R.sup.8 is C.sub.1 -C.sub.6 alkyl, C.sub.2 -C.sub.6 alkenyl, C.sub.3 -C.sub.8 cycloalkyl, phenyl, substituted phenyl, or naphthyl;
- R.sup.10 is C.sub.1 -C.sub.6 alkyl, phenyl or phenyl monosubstituted with halo or C.sub.1 -C.sub.4 alkyl;
- m is 1 or 2;
- and pharmaceutically acceptable salts thereof.

A pharmaceutical formulation comprising an effective amount of a compound of Formula ##STR20## wherein:
- R.sup.1 and R.sup.2 are independently hydrogen, C.sub.1 -C.sub.4 alkyl, benzyl, or .alpha.-methyl-4-nitrobenzyl;
- X is nitro, halo, --OH, --NH.sub.2, --CN, --NHC(O)R.sup.3, --C(O)R.sup.6, --NHSO.sub.2 R.sup.7, or --SO.sub.2 NHR.sup.10 ;
- R.sup.3 is C.sub.1 -C.sub.6 alkyl, C.sub.2 -C.sub.6 alkenyl, C.sub.3 -C.sub.8 cycloalkyl, phenyl, substituted phenyl, naphthyl, or phenyl(C.sub.1 -C.sub.4 alkylene);
- R.sup.6 is hydroxy, amino, C.sub.1 -C.sub.6 alkoxy, benzyloxy, phenoxy, or --NHR.sup.8 ;
- R.sup.7 is C.sub.1 -C.sub.6 alkyl, phenyl or phenyl monosubstituted with halo or C.sub.1 -C.sub.4 alkyl;
- R.sup.8 is C.sub.1 -C.sub.6 alkyl, C.sub.2 -C.sub.6 alkenyl, C.sub.3 -C.sub.8 cycloalkyl, phenyl, substituted phenyl, or naphthyl;
- and
- R.sup.10 is C.sub.1 -C.sub.6 alkyl, phenyl or phenyl monosubstituted with halo or C.sub.1 -C.sub.4 alkyl;
- m is 1 or 2;
- or a pharmaceutically acceptable salt thereof, in combination with a suitable pharmaceutical carrier, diluent, or excipient.
